COX Inhibition Assay
Cyclooxygenase-2 or COX-2 is the enzyme responsible for the synthesis of prostanoids from arachidonic acid. Besides its role in inflammation, it is currently under study for a role in cancer and cardiovascular diseases.
We are offering an assay crucial for the field of drug discovery. We assay the inhibitory effect of new molecules, natural or synthesized, on both COX-1 and COX-2 enzymes.
Principle of the Assay:
The assay relies on the fluorometric detection of prostaglandin G2 formed by the COX enzyme. This gives a huge advantage over the conventional colorimetric determination in being more sensitive and suitable for naturally colored compounds.
Assays available and the expected output:
- Quick Screening:
- One concentration will be tested (100 µg/mL or µM (in case the sample is a pure compound)) to determine the %inhibition of the enzyme at this concentration. The result will be expressed as the % inhibition. If the client requires a specific screening concentration it should be stated.
- IC50 Determination:
- 5 concentrations will be tested to determine the concentration at which 50% of the enzyme is inhibited. The results will be expressed as the value of the IC50 of the sample(s) as well as an IC50 curve.
N.B: In case the value of the IC50 is higher than 100µg/mL (or 100µM for pure compounds), the result will be represented as the % inhibition at this concentration, and the IC50 value will not be calculated.
