Cellular Drug Uptake by HPLC
Cellular drug uptake assays using living cells are central determinants of the pharmacokinetic and pharmacodynamic properties of drugs and drug candidates. They can be used to examine the effects of compounds on transporter or receptor systems.
Applications
Measuring the transport or internalization of biologically active molecules (drugs, nanoparticles,) into cells that are key factors in
- The discovery and development of novel drugs.
- Determination of the compound’s potency and efficacy.
- Limiting the toxic effects and increasing the desired effects.
- Estimating the drug selectivity that often depends on the presence or absence of specific transporters.
Work protocol
Protocol might differ slightly from one experiment to another but it includes 3 main steps:
- Cell treatment: conducted on either normal or cancer cell lines
- Cell lysis & drug extraction
- Drug quantification using HPLC.
