Oxidative stress is injurious to cells causing damage to DNA, proteins and lipids. It is caused by increased levels of reactive oxygen species (ROS) or/and decreased levels of antioxidants including reduced glutathione (GSH). Oxidative stress has been implicated to play a role in ageing and many pathological diseases, including cancer, diabetes, cardiovascular disorders, atherosclerosis, Parkinson’s and Alzheimer’s Diseases.

TNF-α (Tumor Necrosis Factor Alpha) ELISA Assay

Tumor necrosis factor (TNF), a 17 kDa protein composed of 157 amino acids. It is mainly produced by activated T lymphocytes, macrophages, and natural killer (NK) cells. It plays a critical role in immune system for cell signaling. If macrophages detect any infection, immediately TNF is released to alert other immune system cells as part of an inflammatory response.

 

Assay Principle

The micro ELISA plate is pre-coated with an antibody specific to Rat TNF-α. After adding samples/standards, a biotinylated detection antibody specific for Rat TNF-α and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each well and incubated. The optical density of TNF-α conjugated with the biotinylated detection antibody is measured spectrophotometrically at a wavelength of 450 nm. The OD value is proportional to the concentration of Rat TNF- α.
